The inhibitory effect on hydrogen peroxide(H2O2) of rifampin in Macrophage infected with M. leprae

Abstract
According to the previous reports, phagocyted Mycobacterium leprae were killed by
free radicals(H2O2, O2£þ, NO£þ), pH and
enzyme secreted from phagosome-lysosome of macrophages, otherwise progress to the
leprosy. If viability of M. leprae related with free radicals from Macrophages phgocyted
intracellular bacilli(M. leprae), level of H2O2 could indicate
sensitivity of MDT.
We have extimated that relationship between hydrogen peroxide and serially diluted
rifampin with does of 0, 5, 10, 20, 40, 80¥ìg/ml to M. leprae infected IC-21 macrophage.
M. leprae infected IC-21 macrophages exhibited about 7,000 RFU(Relative Fluorescent
Unit) in comparison with control macrophages showing about 5,000 RFU. Rifampin
treatment enhanced low production of H2O2 with does
dependent pattern. H2O2 production of M. leprae infected
macrophage could reflect effectiveness of MDT containing rifampin. Further study
regarding evaluation of viability of M. leprae could confirm our results.
